Dean Robertson's side lead 6½-5½ after a dramatic opening day on the County Clare links, leaving the holders needing 6½ points from Sunday's 14 matches to retain the trophy and the home side requiring seven to reclaim it for the first time since 2015.
The hosts made the stronger start, taking a 2½-1½ advantage from the morning foursomes.
Connor Graham and Tyler Weaver edged Jay Leng and Kihei Akina 1up, while Niall Sheils-Donegan and Josh Hill produced the day's biggest foursomes win, beating Josiah Gilbert and Ryder Cowan 4&3.
Their match also saw Sheils-Donegan involved in a rules blunder.
The other two matches were decided by the narrowest of margins.
Stuart Grehan and Eliot Baker halved with Preston Stout and Ethan Fang, while Miles Russell and Tyler Watts defeated Luke Poulter and Sam Easterbrook 1up.
The United States fought back in the afternoon singles, but could not overturn the deficit.
Grehan produced one of the day's standout performances, recovering from a difficult start to defeat world number one Preston Stout 1up.
The Irishman won the final three holes to complete a remarkable comeback and give GB&I an early boost in the singles.
The Americans responded with wins for Ethan Fang, William Jennings, Jay Leng and Stewart Hagestad, leaving the teams level on points for the afternoon at 4-4.
GB&I, however, finished strongly.
Poulter beat Russell 2&1, Sheils-Donegan defeated Watts 3&2 and Eliot Baker overcame Gilbert by the same margin to ensure the hosts retained their overall advantage.
The result leaves everything to play for on Sunday. Four foursomes will open the final day before all 10 players from each team contest singles, with 14 points available.
GB&I are attempting to end an 11-year wait for the Walker Cup, having last won at Royal Lytham & St Annes in 2015.
The United States, who won the 2025 match at Cypress Point 17-9, are bidding for a sixth successive victory.
