Russell Henley carded a bogey-free nine-under 61 to open up a two-shot lead after 18 holes of the PGA Tour's $40m season finale.
Henley tore round East Lake on Thursday, hitting seven birdies and an eagle to lead four-time major champion and pre-tournament favourite Scottie Scheffler by two strokes.
Scheffler finished with back-to-back birdies to card a 63 and is one stroke clear a group of five players on six-under.
That group includes Collin Morikawa, Justin Thomas, Patrick Cantlay, Robert MacIntyre and Tommy Fleetwood.
MacIntyre surrendered a four-shot lead to the aforementioned Scheffler last week at the BMW Championship.
Three-time FedEx Cup champion Rory McIlroy was noticeably better off the tee, hitting 10 of 14 fairways for a 66.
The Tour Championship is the PGA Tour's season finale, featuring only the top 30 in the FedEx Cup standings.
It is the final tournament before U.S. Ryder Cup captain Keegan Bradley names his six wildcards for the forthcoming showdown at Bethpage Black.
Bradley knows all six of his automatic qualifiers, but he must decide who will join Scheffler, JJ Spaun, Harris English, Xander Schauffele, Henley and LIV Golf recruit Bryson DeChambeau.
The 39-year-old will confirm his selections on 26 August, whilst his European counterpart will reveal his on 1 September.
