Justin Rose shot a four-under par 68 to extend his lead to six shots at the Farmer Insurance Open in San Diego.
The 45-year-old former world number one, who leads America's Joel Dahmen, sits on 21-under par after 54 holes.
Japan's Ryo Hisatsune and South Korea's Si Woo Kim are tied for third on 13-under, with Ireland's Seamus Power and former Masters champion Hideki Mastuyama also in the top-10.
Rose, who broke his own 36-hole scoring record by two shots yesterday, was once again in full control of his game.
Four birdies on the front nine and another at the 10th saw him build an eight-shot advantage.
But dropped shots at the 11th, 12th and 16th gave the chasing pack hope.
Dahmen birdied two of his three final holes to close the gap - but it will take something extremely special on Sunday to stop Rose.
"Today I played well, gave myself a lot of chances," said Rose.
"In some ways, yeah, you walk off like thinking maybe it could have been lower, you know.
"You never want to say that after a 68 at Torrey or, you know, obviously being in the position that I'm in, but yeah I played really, really well today.
"Like I thought that was the round of the week from tee to green anyway."
Rose vowed not to get complacent.
"I think there's always enough respect for the game of golf in the back of your mind that you've got to do everything right tomorrow," he said.
"You're going to come out, have to be focused, have to play well.
"Obviously somebody can always really shoot a great round and therefore you've got to get around this golf course in a pretty decent score."
Elsewhere, five-time major champion Brooks Koepka struggled to climb the leaderboard on moving day.
The former LIV recruit made the cut on the number, but laboured to a one-over 73 on Saturday.
Koepka made five birdies, four bogeys and one double-bogey.
"[I am] playing well," said Koepka.
"[But] putting, I've never felt comfortable on poa. You just miss a few and you've got zero confidence.
"I don't know, today I felt every three, four-footer was slightly downhill and I felt like it kind of went a little bit of both ways.
"I tried to firm 'em all and take the break out to still be aggressive like I normally am, but I wasn't even hitting the hole.
"I don't know, I'm just going to chalk it up to just a bad day, but I'm happy with everything else."
Several big names missed the cut on Friday. Xander Schauffele's streak of 72 - the longest active on the PGA Tour - came to an end.
Patrick Cantlay, Will Zalatoris, J.J. Spaun, Max Homa, Marco Penge and Ludvig Aberg also missed the cut.
