The PGA Tour has confirmed to GolfMagic that Koepka's withdrawal is due to a personal reason.
Koepka's withdrawal gives him an uphill battle to make next month's FedEx Cup Playoffs.
The 36-year-old was American was forced to withdraw before the final round of last month's RBC Canadian Open because of a hand injury, which appeared to scupper his chances of contending for the season's final two majors.
The former World No.1 was unable to build any momentum on his return, missing the cut at both the U.S. Open and Genesis Scottish Open before producing a more encouraging display at The Open, where he finished tied for 28th at Royal Portrush.
Now ranked 84th in the FedEx Cup standings, Koepka's latest withdrawal deals another blow to his hopes of extending his PGA Tour season.
With only next week's Wyndham Championship remaining before the top 70 qualify for the FedEx St. Jude Championship, time is running out for the LIV Golf star to climb back into Playoff contention.
The PGA Tour has confirmed Ben Silverman will replace Koepka in the Rocket Classic field.
Silverman currently sits 134th in the FedEx Cup standings.
The top 70 players following next week's Wyndham Championship will advance to the FedEx St. Jude Championship, the opening event of the FedEx Cup Playoffs.
Koepka is not the only notable absentee from this week's tournament.
Jason Day also withdrew from the Rocket Classic on Monday due to a recurring back injury, marking the second consecutive PGA Tour event he has been forced to miss.
The Australian also pulled out of last week's 3M Open because of the same issue and has now withdrawn from consecutive tournaments as he battles another setback in a career-long struggle with back problems.
Day, a 13-time PGA Tour winner and the 2015 PGA Championship winner, has dealt with fitness concerns throughout much of his career but had appeared to put those issues behind him in recent seasons.
However, the 38-year-old was forced to withdraw after just 10 holes of last month's U.S. Open because of the same back complaint.
He returned to finish tied for 55th at the Travelers Championship before missing the cut at The Open, but has been unable to compete since.
Day's latest withdrawal is another blow to his FedEx Cup Playoff hopes.
He slipped to 74th in the standings following last week's 3M Open withdrawal and now faces an uphill battle to break back into the top 70 before the regular season concludes.
The PGA Tour has confirmed Hayden Springer has replaced Day in the Rocket Classic field.
Springer is lying 132nd in the FedEx Cup standings on the eve of this week's tournament in Michigan.
Detroit Golf Club will host its final ever Rocket Classic this week.
